Clinical Pharmacology of COX-2 Inhibitors

By Suzanne Holm, MS, ARNP, CS

Clinical trials have shown that COX-2 inhibitors are equivalent in efficacy to nonselective NSAIDs and have a lower incidence of GI toxicity. This interactive course reviews the clinical pharmacology of COX-2 inhibitors and discusses their role in pain management and in neurodegenerative disease.

Overview

Traditional non-steroidal anti-inflammatory drugs (NSAIDs) are generally well-tolerated but they do cause adverse GI effects in some patients. Because these agents are so widely used, the aggregate morbidity and mortality resulting from their adverse effects is significant.

Cyclooxygenase-2 (COX-2) inhibitors constitute a new group of NSAIDs which block prostaglandin production by cyclooxygenase-2, but not by cyclooxygenase-1. (Traditional NSAIDs block both.) Clinical trials have shown that COX-2 inhibitors are equivalent in efficacy to nonselective NSAIDs and have a lower incidence of GI toxicity.

In this interactive course we will review the clinical pharmacology of COX-2 inhibitors and discuss the role of COX-2 inhibitors in pain management and in neurodegenerative disease.
